Genetic Map of Bacteriophage 4 X 174

چکیده
Bacteriophage 46X174 temperature-sensitive and nonsense mutations in eight cistrons were mapped by using two-, three-, and four-factor genetic crosses. The genetic map is circular with a total length of 24 X 104 wt recombinants per progeny phage. The cistron order is D-E-F-G-H-A-B-C. High negative interference is seen, consistent with a small closed circular deoxyribonucleic acid molecule as a genome.

Endogenous transmembrane tunnel formation mediated by phi X174 lysis protein E.
Biochemical and genetic studies have suggested that a transmembrane tunnel structure penetrating the inner and outer membranes is formed during the lytic action of bacteriophage phi X174 protein E. In this study we directly visualized the lysis tunnel by using high-magnification scanning and transmission electron microscopy.

Mycoplasmavirus MVL2 is a nonlytic enveloped virion containing DNA. This DNA has been shown to be a double-stranded circular superhelical molecule of 11.8 kilobase pairs (7.8 X 10(6) daltons). The superhelix density is greater than that of phi X174 RFI but less than that of PM2 phage DNA. A physical map of the MVL2 genome has been obtained using restriction endonucleases.
متن کاملImmune response to phi X 174 in man. 5. Primary and secondary antibody production in primary biliary cirrhosis.
In primary biliary cirrhosis, the primary immune response to the bacteriophage phi X 174 is normal but the secondary response is significantly reduced. The reduction is primarily of IgG antibody, while IgM is proportionately less affected. These changes may be the result of a reduction in helper T lymphocyte function and may contribute to the increase in the ratio of serum concentration of IgM ...
متن کاملGene D of bacteriophage phi X 174: absence of transcriptional and translational regulatory properties.
The only evidence that a gene product of phi chi 174 is involved in the regulation of phage transcription was presented by Clements and Sinsheimer (1974) for gene D. I. found, in attempting to investigate this possibility further, that the D protein had no regulatory effect. When the D protein was eliminated by various nonsense mutations, all other phage proteins were synthesized at rates nearl...
متن کاملReplication of bacteriophage phiX174 in a temperature-sensitive deoxyribonucleic acid host cell.
Bacteriophage phiX174 is unable to replicate in Escherichia coli t3 at the restrictive temperature. However, if progeny phage synthesis is initiated at the permissive temperature, it will continue after a shift to the restrictive temperature.
